RESUMO

T cells are broadly categorized into two groups, namely conventional and unconventional T cells. Conventional T cells are the most prevalent and well-studied subset of T cells. On the other hand, unconventional T cells exhibit diverse functions shared between innate and adaptive immune cells. During recent decades, γδ T cells have received attention for their roles in cancer immunity. These cells can detect various molecules, such as lipids and metabolites. Also, they are known for their distinctive ability to recognize and target cancer cells in the tumor microenvironment (TME). This feature of γδ T cells could provide a unique therapeutic tool to fight against cancer. Understanding the role of γδ T cells in TME is essential to prepare the groundwork to use γδ T cells for clinical purposes. Here, we provide recent knowledge regarding the role γδ T cell subsets in different cancer types.

Indoleamine-2,3 dioxygenase is a rate-limiting enzyme in the tryptophan catabolism in kynurenine pathways that has an immunosuppressive effect and supports cancer cells to evade the immune system in different cancer types. Diverse cytokines and pathways upregulate the production of indoleamine-2,3 dioxygenase enzymes in the tumor microenvironment and cause more production and activity of this enzyme. Ultimately, this situation results in anti-tumor immune suppression which is in favor of tumor growth. Several inhibitors such as 1-methyl-tryptophan have been introduced for indoleamine-2,3 dioxygenase enzyme and some of them are widely utilized in pre-clinical and clinical trials. Importantly at the molecular level, indoleamine-2,3 dioxygenase is positioned in a series of intricate signaling and molecular networks. Here, the main objective is to provide a focused view of indoleamine-2,3 dioxygenase enhancer pathways and propose further studies to cover the gap in available information on the function of indoleamine-2,3 dioxygenase enzyme in the tumor microenvironment.

Cancer is classified into metabolic and/or genetic disorders; notably, the tryptophan catabolism pathway is vital in different cancer types. Here, we focused on the interaction and molecular connection between the cytotoxic T lymphocyte-associated antigen-4 (CTLA-4) receptor and indoleamine-2,3-dioxygenase (IDO) enzyme. To test the impact of the selected immunotherapies on breast cancer cell migration and cell survival, we used in vitro assays. Also, we test the impact of anti-CTLA-4 antibody on the IDO-positive cells. The results of cell migration and clonogenic assays showed that anti-CTLA-4 antibody reduces cancer cell migration and clonogenic abilities of murine breast cancer cells. In addition, the result of flow cytometry showed that the anti-CTLA-4 antibody did not change the percentage of IDO-positive cancer cells. Notably, administrating an IDO blocker, 1-Methyl-DL-tryptophan (1MT), reduces the efficiency of the antiCTLA-4 antibody. The enzymatic blocking of the IDO reduces the efficiency of the anti-CTLA-4 antibody on cell migration and clonogenic abilities suggesting that there is an inhibitory interaction at the molecular level between functions of CTLA-4 and IDO. It is unclear via which mechanism(s) IDO interacts with CTLA-4 signaling and also why blocking IDO makes disruption in CTLA-4 signaling in cancer cells. Indeed, evaluating the role of IDO in CTLA-4 signaling in cancer cells may assist in clarifying a poor response to CTLA-4 immunotherapies by some patients. Hence, further investigation of the molecular interaction between CTLA-4 and IDO might help to improve the efficiency of CTLA-4 immunotherapy.

In the tumor microenvironment, the function of T cells is a fate-changer for tumor progression. In the meantime, CD28 and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) are vital role players in the controlling activity of T cells as an activator and deactivator, respectively. In T cells in comparison to CD28, the molecular mechanism of CTLA-4 is unclear. In addition, despite the fact that most tumor cell types express CTLA-4, its role in tumor cells is not well understood and only few studies focused on the role of CTLA-4 signaling in tumor cells. It is illustrated that CTLA-4 signaling causes PD-L1 expression in tumor cells. However, numerous characteristics of CTLA-4 signaling in tumor cells are ambiguous and require to be described. In this article, we proposed that the CTLA-4 signaling during immunotherapy with anti-CTLA-4 antibodies may cause poor responses by patients. In addition, we attract attention to several fundamental questions regarding CTLA-4 signaling in tumor cells. Overall, the CTLA-4 signaling function and the related gaps about its role in tumor cells in the present review are challenged.

Kidney diseases are the most common illness for cats with a prevalence seven times higher than in dogs. Metanephros is the last of three renal systems to be formed during the embryonic period, which then becomes the permanent kidney. The current work aimed to analyse the morphology and to quantify the structures present in the development of metanephros from domestic cat (Felis catus) embryos and foetuses. For this purpose, the evaluation of the biometric parameters of metanephros from cat embryos and foetuses was performed in addition to the quantification of renal corpuscles and volume of cortical and medullary layers by stereological analysis. The evaluated biometric parameters were weight, width, height, thickness and volume. The values of the measured biometric parameters increased throughout the gestational stages. The quantity of renal corpuscles gradually increased following the embryo-foetal development, mainly during the middle of the gestational stage. It was during this phase that morphologically, a complete corticomedullary division was observed. Although the difference in the quantity of renal corpuscles between the middle and the end of the gestational stages was not statistically significant, there was an increase in the volume of the medullary layer and a decrease in the volume of the cortical layer between these two stages. These findings suggest that the metanephros presents a progressive growth with the renal corpuscles following this development until the middle of the gestational stage. Starting from this phase, the differentiation of the corticomedullary layers can be seen with a significant increase in the medullary layer.

Germ cell transplantation and testis graft represent promising biotechnologies that can be applied for the reproduction of commercial or endangered species. However, mechanisms of rejection from the host immune system might remove the transplanted donor cells/tissues and limit the surrogate production of gametes. In this work, we administered emulsion containing-immunosuppressants to verify whether they are capable to prevent immune rejection and promote survival of testis allografts in rainbow trout. In the first part of this study, we demonstrated in vitro that tacrolimus and cyclosporine were able to affect viability, inhibit leucocyte proliferation, and suppress il2 expression in vitro. In in vivo experiments, both doses of tacrolimus (0.5 and 1.5 mg/kg) and the lower dose of cyclosporine (20 mg/kg) significantly inhibited the expression of il2 in head kidney, three days post-injection. A higher dose of cyclosporine (40 mg/kg) was able to inhibit il2 expression for up to seven days post-injection. In the second part, testis allografts were conducted in fish treated weekly with emulsion containing-tacrolimus. Immunohistochemical, conventional histology, and qRT-PCR (vasa) analysis demonstrated the presence of spermatogonial cells by the fifth week, in animals treated with 0.5 mg/kg of tacrolimus similar as found in autografted group. In the group treated with the highest tacrolimus dose (1.5 mg/kg) and in the non-treated group (without immunosuppressant), no germ cells or their respective markers were detected. il2 expression in head kidney was also suppressed in grafted animals treated with tacrolimus compared to non-treated group. These results suggest that tacrolimus may be a promising immunosuppressant for testis allografts or germ cell transplantation in rainbow trout. Co-administration combining tacrolimus (at lower dose) with other immunosuppressive drugs for inhibiting other activation pathways of the immune system, as performed in human organ transplantation, could be an alternative approach to optimize the immunosuppressive effects in host organisms.

Indoleamine 2 3-dioxygenase (IDO) is a protein usually described in mammals, which, among other functions, participates in the maternal-fetal tolerance process. The blue-shark, Prionace glauca (Linnaeus, 1758) is a viviparous placentary species in which the yolk sac develops during the pregnancy, turning into a placenta for matrotrophic nutrition of the embryo. The purpose of this study was to investigate the expression of IDO in the P. glauca maternal-fetal interface along three gestation phases and describe its distribution and the meaning of its presence. The results showed IDO labelling during the yolk sac/placenta development in the ectoderm on the three development phases and in the endoderm at the two first phases. In the uterine epithelium, IDO was observed in the last two phases. These interface tissues are major contact areas between the mother and the semiallogeneic conceptus and this relation could induce an immunological response against the fetus. Therefore, the presence of IDO may indicate that it could have a similar role in the mechanism of maternal-fetal tolerance in the P. glauca placental interface, as described in eutherian mammals.

PROBLEM: Several mechanisms contribute to the tolerogenic state observed during pregnancy, such as the activity of the enzyme indoleamine 2, 3-dioxygenase (IDO). This initializes the catabolism of tryptophan, inducing T cells to apoptosis due to its deprivation and by the action of its catabolites in the placental microenvironment. Progesterone plays an important part on immunological tolerance mechanisms during pregnancy; however, there is no evidence it is related to IDO activity. Thus, this study aimed to investigate progesterone influence on the maternal-fetal interface of pregnant Wistar rats, by identifying IDO positive cells by immunophenotyping and flow cytometry under exogenous progesterone supplementation. METHOD OF STUDY: Placenta and embryo cells were cultured and separated into groups that received interferon γ or progesterone, supplemented or not with mifepristone. After 2 and 24 h, these were labeled with an anti-IDO and a series of antibodies specific to leucocytes and progesterone receptor and processed through flow cytometry analysis. RESULTS: Progesterone induced a significant decrease in the expression of IDO in dendritic cells and CD4+ lymphocytes. CONCLUSION: The blocking of progesterone receptor on these cells by mifepristone restored IDO expression levels and may constitute evidence of the participation of this hormone through a direct route in these cells.

This study aimed to analyze the effects of caloric restriction on aged femoral articular cartilage of Wistar rats. Three groups of eight animals each were considered: young (YC) and old (OC) control groups fed with a normal diet and old caloric restriction group (OCR) composed of 18-month-old animals fed with a 31% less caloric diet from 6-months of age. Articular cartilage was studied through morphometry and immunohistochemistry. Body mass was 12% less in the OCR group than in the OC group. The articular cartilage from OC rats show thinner medial condyles, fewer chondrocytes, smaller chondrocytes nuclear volume and, in both condyles, a predominance of collagen type II and less collagen density compared to both YC and OCR groups (p < .001). In contrast, OCR articular cartilage show thicker medial condyles, larger chondrocytes nuclear volume and increased collagen density compared to OC group (p < 0.001). We concluded that caloric restriction minimizes the effects of aging on medial condyles of the femoral articular cartilage.

Sensory organs in elasmobranchs (sharks, skates, rays) detect and respond to a different set of biotic and/or abiotic stimuli, through sight, smell, taste, hearing, mechanoreception and electroreception. Although gustation is crucial for survival and essential for growth, mobility, and maintenance of neural activity and the proper functioning of the immune system, comparatively little is known about this sensory system in elasmobranchs. Here we present a preliminary investigation into the structural and dimensional characteristics of the oral papillae and denticles found in the oropharyngeal cavity of the blue shark (Prionace glauca) during embryonic development through adulthood. Samples were obtained from the dorsal and ventral surface of the oropharyngeal cavity collected from embryos at different development stages as well as from adults. Our results suggest that development of papillae occurs early in ontogeny, before the formation of the oral denticles. The diameter of oral papillae gradually increases during development, starting from 25 µm in stage I embryos, to 110 µm in stage IV embryos and 272-300 µm in adults. Embryos exhibit papillae at early developmental stages, suggesting that these structures may be important during early in life. The highest density of papillae was observed in the maxillary and mandibular valve regions, possibly related to the ability to identify, capture and process prey. The oral denticles were observed only in the final embryonic stage as well as in adults. Accordingly, we suggest that oral denticles likely aid in ram ventilation (through reducing the hydrodynamic drag), to protect papillae from injury during prey consumption and assist in the retention and consumption of prey (through adhesion), since these processes are only necessary after birth.

Lymphocytes and macrophages are the main white cells involved in fetal-maternal tolerance. Little is known about these leukocytes in bovine placenta, such as the quantity and location of these cells. Thus, the objective of this study was to identify lymphocyte and macrophage populations in bovine placenta using specific markers and flow cytometry. This study analyzed samples of placentomes and intercaruncular regions of cows in the three quarters of pregnancy. In the placentomes, during the first quarter of pregnancy, mean percentage of labeled CD3+ cells was 2.34%; CD8+, 1.28%; CD14+, 1.66%; and CD335+, 0.96%. For the intercaruncular region, percentage of CD3+ cells was 0.71%; CD8+, 1.63%; CD14+, 2.81%; and CD335+, 2.81%. In the second quarter, placentomes showed 0.94% CD3+ cells; 0.77% CD8+; 0.72% CD14+; and 0.51% CD335+. In the intercaruncular region, percentage of labeled CD3+ cells was 0.50%; CD8+, 1.81%; CD14+, 2.64%; and CD335+, 0.51%. In the third quarter, placentomes showed labeling of 0.88% CD3+; 0.66% CD8+; 1.06% CD14+; and 0.74% CD335+ cells. In the intercaruncular region, percentage of labeled CD3+ cells was 0.19%; CD8+, 2.23%; CD14+, 2.43%; and CD335+, 0.16%. The results showed that there was a greater immunomarking of leukocytes CD3+ and CD335+ in the placentome when compared to the intercaruncular region during the third trimester. It can be concluded that leukocytes populations in bovine placenta is reduced, probably because of the syndesmochorial characteristic of bovine placenta. This represents a significant barrier for the immunological system of the mother, sharply decreasing the exposure of the conceptus to the mother's immune system.(AU)


Linfócitos e macrófagos são os principais leucócitos envolvidos na tolerância materno-fetal. Pouco se sabe sobre esses leucócitos na placenta bovina, como por exemplo, a quantidade e localização dessas células. Assim, o objetivo desse estudo foi identificar populações de linfócitos e macrófagos na placenta bovina utilizando marcadores específicos e citometria de fluxo. Este estudo analisou amostras de placentônios da região intercaruncular de bovinos nos três trimestres da gestação. No primeiro trimestre, nos placentônios, a porcentagem média de células CD3+ foi 2,34%; CD8+, 1,28%; CD14+, 1,66%; e CD335+, 0,96%. Na região intercaruncular, a porcentagem de células CD3+ foi 0,71%; CD8+, 1,63%; CD14+, 2,81%; e CD335+, 2,81%. No segundo trimestre, os placentônios apresentaram 0,94% de células CD3+; 0,77% de CD8+; 0,72% de CD14+e 0,51% de CD335+. Na região intercaruncular, a porcentagem de células CD3+ foi 0,50%; CD8+, 1,81%; CD14+, 2,64%; e CD335+, 0,51%. No terceiro trimestre, os placentônios apresentaram 0,88% de células marcadas CD3+; 0,66% de CD8+; 1,06% de CD14+ e 0,74% de CD335+. Os resultados mostraram que a imunomarcação de leucócitos na região do placentônio foi maior do que na região intercaruncular no terceiro trimestre. Concluiu-se que a população de leucócitos CD3+ e CD335+ na placenta bovina está reduzida, provavelmente devido à sua característica sindesmocorial. Essa característica representa uma barreira significante para o sistema imunológico da mãe, o que diminui drasticamente a exposição do concepto ao sistema de defesa da mãe.(AU)

Some batoid species are covered with dermal denticles (or placoid scales) that occasionally develop into thorns. In sexually mature males, sharp teeth and alar thorns found on the apex of the lateral disc are used to hold the female during copulation. This study set out to analyze microscopic features of modified dermal denticles and thorns and to investigate sexual dimorphism in Atlantoraja cyclophora and A. castelnaui species. Skin samples collected from areas covered with thorns were fixed in 10% formaldehyde, processed and analyzed using scanning electron microscopy. Alar thorn morphology varied within species, while caudal thorn, rostral and caudal dermal denticle morphology varied within and between species. These structures play an important role in the protection and reproduction of the species studied and constitute important taxonomic information, given they are often the only elements preserved in archaeological sites and fossil records.

A indoleamina 2,3-dioxigenase (IDO) é uma enzima que cataboliza o aminoácido triptofano, levando à inibição da proliferação de linfócitos T, seja pela exaustão desse aminoácido no ambiente, ou pela indução via catabólitos induzindo-os a apoptose. Em mamíferos, esta enzima atua em diversas condições do organismo como a gestação, infecções, inflamações crônicas, transplantes e tumores, atuando na regulação imunológica. Estudos recentes identificaram a presença de moléculas homólogas a IDO em espécies filogeneticamente inferiores, cuja função parece estar restrita ao metabolismo do triptofano como fonte de energia. Este estudo teve por objetivo averiguar a expressão da IDO em células sanguíneas e órgãos hematopoiéticos de truta arco-íris pela imuno-histoquímica, buscando evidências de que a mesma poderia, nesta espécie, estar relacionada ao sistema imune. A expressão de IDO foi observada nos órgãos hematopoiéticos estudados incluindo o rim cefálico que apresentou marcação em células interrenais e leucócitos; baço, na qual a marcação restringiu à alguns leucócitos; no fígado a marcação ficou limitada à apenas algumas células dentro dos vasos sanguíneos e nas extensões sanguíneas pode-se visualizar a marcação de alguns leucócitos como os monócitos, linfócitos e neutrófilos. A predominância da marcação da IDO nesses tecidos pode constituir uma evidência de que a IDO identificada na O. mykiss esteja relacionada ao sistema imunológico nessa espécie...


Indoleamine 2,3-dioxygenase (IDO) is an enzyme that catabolizes the amino acid tryptophan, leading to inhibition of T lymphocyte proliferation, whether by depletion of this amino acid in the environment, or by induction via the catabolites inducing apoptosis. In mammals, this enzyme acts on various conditions of the body such as pregnancy, infections, chronic inflammation, transplantation and tumors, acting in immune regulation. Recent studies have identified the presence of homologous molecules IDO lower phylogenetically related species, whose function appears to be confined to the tryptophan metabolism as an energy source. This study aimed to investigate the expression of IDO in blood cells and hematopoietic organs of rainbow trout by immunohistochemistry, seeking evidence that it could, this species is related to the immune system. The expression of IDO was observed in hematopoietic organs studied including head kidney that show labeling in interrenal cells and leukocytes; spleen, in which the marking restricted to a few leukocytes in the liver;, labeling was restricted to only certain cells within the blood vessels and the blood extensions can view the marking of some leukocytes including monocytes, lymphocytes and neutrophils. The predominance of IDO marking these tissues may constitute evidence that IDO identified in O. mykiss is related to the immune system in this species...

A indoleamina 2,3-dioxigenase (IDO) é uma enzima responsável por catabolizar o aminoácido triptofano. Sua presença no ambiente uterino placentário está relacionada à tolerância imunológica ao semi-aloenxerto, pois impede a proliferação de células imunológicas maternas, seja pela falta do aminoácido, ou pela ação de alguns catabólitos oriundos da quebra do triptofano, como o ácido quinolínico, que é tóxico principalmente para os linfócitos T. Pouco se conhece sob a influência de substâncias (hormônios e citocinas) presentes na interface materno fetal e a expressão dessa enzima. Por esta razão, formulou-se a hipótese de que hormônios e interleucinas presentes na região uteroplacentária poderiam exercer algum efeito na expressão da IDO. Células oriundas da interface materno fetal de ratas Wistar foram mantidas em cultivo, onde receberam suplementação com estradiol e interferon-γ. A expressão da enzima foi avaliada pela técnica de citometria de fluxo nos períodos de 4, 24 e 48 horas e confirmação da presença proteica por imuno-histoquímica. Os resultados mostraram um aumento na expressão de IDO após a adição de estrógeno (9,03±0,81/11,25±0,25) e interferon-γ (9,03±0,81/20,43±0,60). O efeito do interferon-γ já era esperado como relatado na literatura, contudo, a elevação da expressão da IDO pela adição do estrógeno constitui nova informação sobre possíveis mecanismos envolvidos na ativação da enzima. O melhor esclarecimento desses achados poderia contribuir para uma melhor compreensão da participação dessa enzima na tolerância materno-fetal e para uma futura modulação terapêutica da mesma...


The indoleamine 2,3-dioxygenase (IDO) is an enzyme responsible for catabolizing the tryptophan. Its presence in the placental uterine environment is related to immunological tolerance to the semi-allograft because it prevents proliferation of maternal immune cells, either by the lack of this amino acid or by the action of its catabolites, such as the quinolinic acid, which is particularly toxic for T lymphocytes. Little is known regarding the influence of hormones and cytokines on the expression of IDO in the maternal fetal interface. Therefore, the hypothesis that some hormones and interleukins present in uteroplacental region could have an effect on the expression of IDO on cultured cells was tested. Cells derived from the fetal maternal interface from Wistar rats were kept in culture and supplemented with estradiol and interferon-γ. Expression of the enzyme was assessed by flow cytometry at periods of 4, 24 and 48 hours and confirmation of the presence of protein by immunohistochemistry. The results showed an increasing of IDO expression after the addition of estrogen (9.03±0.81 to 11.25±0.25) and interferon-γ (9.03±0.81 to 20.43±0.60). The effect of interferon-γ was expected as reported in the literature, however, elevated IDO expression by estrogen represents new information on possible mechanisms involved in the enzyme activation. These findings could provide a better understanding of IDO contribution on maternal-fetal tolerance and may collaborate to future therapeutic modulation of this enzyme...

O sistema respiratório das aves é bastante eficiente com pulmões pequenos e compactos ligados a sacos aéreos que diferem entre as espécies. Dessa forma, objetivou-se realizar a descrição anatômica dos sacos aéreos em patos em relação à sua topografia. Para tanto, foram utilizados o trato respiratório pós-cefálico de quatro patos adultos, machos e fêmeas, injetados, via sondagem traqueal, com látex corado e fixados em solução de formol a 10%. Observou-se a presença dos sacos aéreos cervical, clavicular, torácico cranial, torácico caudal e abdominal. O saco aéreo cervical distribuiu-se nos dois antímeros, na região dorsal, localizando-se dorso-lateralmente a musculatura do pescoço. O saco aéreo clavicular apresentou-se com uma distribuição irregular, formando uma porção mediana e duas laterais com inúmeras projeções entre os órgãos das regiões cervical e torácica cranial, apresentando divertículos para os ossos coracóide, úmero e esterno. Sua porção mediana formou, em alguns casos, uma projeção ao redor do coração em "forma de saia". Os sacos aéreos torácicos craniais encontraram-se caudalmente ao coração, medialmente as quatro primeiras costelas, sendo sobrepostos pelo fígado. Os sacos aéreos torácicos caudais localizaram-se em uma posição média aos sacos aéreos torácico cranial e abdominal, estendendo-se além da margem da última costela, englobando a porção cranial do saco aéreo abdominal. Os sacos aéreos abdominais encontraram-se caudo-medialmente aos sacos aéreos torácicos caudais e lateralmente ao trato digestório. Os sacos aéreos torácicos e abdominais apresentaram poucas projeções entre os órgãos, possuindo margens regulares e grande capacidade de volume.


The respiratory system of birds is very effective, comprised by small and compact lungs associated with air sacs that differ between species regarding their topography. Considering this latter aspect, this work aimed to perform an anatomical and topographical description of the air sacs in mallard (Anas platyrhynchos). Ten mallards, five males and five females, had theirpost-cephalic respiratory tract injected with colored latex and fixed by a 10% formaldehyde solution. The presence of cervical, clavicular, cranial thoracic, caudal thoracic and abdominal air sacs was observed. The cervical air sac was distributed in both antimeres situated dorso-laterally to the muscles of the neck. The clavicular air sac presented an irregular distribution, showing a middle and two lateral parts with numerous projections between the cervical and cranial thoracic organs and presenting several diverticula to the coracoids , sternum and humerus bones. Its middle portion formed, in some cases, a projection around the heart. The cranial thoracic air sacs were found caudal to the heart, medially to the first four ribs, being overplayed by the liver. The caudal thoracic air sacs were located in a middle position to the cranial thoracic and abdominal air sacs extending beyond the edge of the rib, over the cranial portion of the abdominal air sac. The abdominal air sacs were found caudal-medial to the caudal thoracic air sacs and lateral to the digestive tract. The thoracic and abdominal air sacs presented few projections between the organs, has and showed regular margins and a high volumetric capacity.

A descrição anatômica articular evidencia os fatores padrões para as condições biomecânicas que devem ser consideradas na seleção do animal de uma determinada espécie. Dessa forma, objetivou-se descrever a articulação metacarpo-falângica na espécie ovina, em relação a sua arquitetura, considerando o volume, recessos, comunicações e relações no aspecto anatômico macroscópico. Para tanto, foram utilizados dez animais adultos da espécie ovina, raça Santa Inês, machos e fêmeas, escolhidos aleatoriamente. A cápsula articular teve uma dimensão média de 3,05 cm e uma capacidade volumétri-ca média de 3,99 ml; as comunicações ocorreram em um grau médio de expansão do Látex nas faces dorsal (56,7%) e palmar (53,3%); as comunicações proximais nas faces dorsal, palmar e dorso-palmar apareceram em 100,0% dos casos; os recessos estiveram presentes nas extremidades distais. Nesse contexto, conclui-se, que a articulação metacarpo-falângica de ovinos apresenta-se com relações topográficas semelhantes à descrita para os demais ruminantes, possuindo comunicações em sua porção proximal e recessos distalmente.


The joints anatomical description highlights the standards for biomechanical conditions that must be considered in the selection of a particular animal species. Thus, the objective was to describe the metacarpal phalangeal joint in sheep, for its architecture and adjacent structures. For this purpose, we used ten adult sheep, Santa Ines breed, males and females, randomly chosen. The anatomical specimens for the study were collected by the disarticulation of the distal row of carpal and metacarpal bone of an sheep's forelimb. To observe the joint capsule, adjacent structures were overruled and their topographical relationships with such joint analyzed. The longitudinal measures of the joint capsule and the III-IV metacarpal bones, proximal phalanges III and IV proximal phalanx were taken. Through the results, it is concluded that the metacarpal phalangeal joint in sheep presents similar topographical relationships described for other ruminants, with connections at the proximal and recesses at the distal areas.

RESUMO

The neurohistologic observations were performed using the specimens prepared by Winkelmann and Schmitt silver impregnation method. The tissues were fixed in 10 percent formalin solution and sections of 40µm thickness were obtained by Leica Cryostat at -30ºC. The sections of dorsal mucosa of White-lipped peccary tongue showed numerous filliform and fungiform papillae, and two vallate papillae on the caudal part. The epithelial layer revealed queratinized epithelial cells and the connective tissue papillae of different sizes and shapes. Thick nerve fiber bundles are noted into the subepithelial connective tissue of the papillae. The connective tissue of fungiform and vallate papillae contained numerous sensitive nerves fibers bundles forming a complex nerve plexus.


As observações neuro-histológicas foram realizadas utilizando amostras preparadas segundo o método de impregnação por prata de Winkelmann e Schmitt. Os tecidos foram fixados em solução de formol a 10 por cento e seções de 40µm de espessura foram obtidas em criostato Leica -30ºC. As seções da mucosa dorsal da língua de queixada revelaram numerosas papilas filiformes, fungiformes e duas papilas valadas sobre a parte caudal. A camada epitelial revelou células epiteliais queratinizadas e papilas de tecido conjuntivo de diferentes tamanhos e formas foram observadas. Espessos feixes de fibras nervosas são notados no tecido conjuntivo subepitelial das papilas. O tecido conjuntivo das papilas fungiformes e valadas contêm numerosos feixes de nervos de fibras sensíveis formando um plexo nervoso complexo.

Avaliou-se, por citometria de fluxo, a fagocitose de Staphylococcus aureus conjugados com iodeto de propídio (IP), por leucócitos circulantes obtidos de cinco fêmeas bovinas negativas no sorodiagnóstico para a Leucose Enzoótica Bovina (LEB); de cinco fêmeas infectadas, manifestando linfocitose persistente (LP); e de cinco fêmeas infectadas, porém alinfocitóticas. Observou-se que, entre as amostras dos animais soronegativos, a porcentagem média de células realizando fagocitose (12,90%) não diferiu da observada entre as células dos animais alinfocitóticos (14,70%). Contudo, ambas foram maiores (p=0,047) que aquela verificada entre as células obtidas de animais manifestando LP (7,20%). Além disso, a intensidade média de fagocitose (caracterizada pela intensidade de fluorescência do IP, em valores arbitrários), verificada em leucócitos de animais manifestando LP (17,43) foi menor (p<0,001) que a observada em leucócitos de animais alinfocitóticos (29,50), e que a observada em leucócitos de animais soronegativos (25,18), que não diferiram entre si. Assim, os resultados permitem-nos alvitrar que há alteração na função fagocítica de leucócitos circulantes em animais infectados pelo vírus da LEB, manifestando LP.


This study evaluated the phagocytosis of propidium iodide-labeled Staphylococcus aureus (PI-Sa) by circulating leukocytes obtained from five Enzootic Bovine Leukosis (EBL)-negative cows, five naturally EBL-infected, non-lymphocytotic cows, and five EBL-positive cows with persistent lymphocytosis (PL), analyzed by flow cytometry. Among cells obtained from EBL-infected cows, presenting PL, the percentage of leukocytes carrying out phagocytosis (7.20%), was smaller (p=0.047) than that verified among cells obtained from non-infected (12.90%), and from BLV-infected, non-lymphocytotic cows (14.70%). Furthermore, leukocytes obtained from EBL-infected cows, presenting PL, showed smaller phagocytosis intensity (characterized by the intensity of propidium iodide fluorescence) than leukocytes obtained from non-infected and from EBL-infected, non-lymphocytotic, cows (p<0.001). Therefore, results show a decreased phagocytic function among circulating leukocytes obtained from BLV-infected, lymphocytotic cows.

A distrofia muscular dos cães Golden Retriever (GRMD), uma miopatia degenerativa causada pela ausência da distrofina é geneticamente homóloga à distrofia muscular de Duchenne que acomete humanos, portanto, estes cães são considerados modelos experimentais para estudos em terapia celular. Seu sucesso depende da imunossupressão adequada. A ciclosporina A (CsA) é indicada para tal e a monitorização de suas concentrações sangüíneas e efeitos adversos são essenciais para viabilizar a terapia. Foram estudados cães GRMD, e normais da mesma raça, submetidos a terapia com CsA, associada, nos GRMD, ao transplante de células tronco. Foram avaliados as concentrações sangüíneas do fármaco através de amostras coletadas a cada dois ou três dias e analisadas pelo método do imunoensaio por fluorescência (FPIA). Como resultado observamos que as concentrações de CsA oscilaram muito, em seis dos oito animais. Concluímos que as doses variam individualmente sendo de maior importância avaliar a concentração do fármaco no sangue e sua viabilização no uso da terapia celular


The muscular dystrophy of Golden Retriever (GRMD) is a degenerative miopaty caused by the absence of dystrophy and it is genetically homologue of the Duchenne muscular dystrophy in humans, so, these dogs are considerably experimental models for studies on cellular therapy. Their successful depends of the adequate immunosuppression. Cyclosporin A (CsA) is indicated for that and the monitoring of the blood concentration and adverse effects are essential to viabilise the therapy. It was studied GRMD dogs, and normal dogs from the same breed, submitted for therapy with CsA, associated, on GRMD, of cell transplantation. It was evaluated blood concentration of the drug, between two or tree days using the method of FPIA. In our results we found that the CsA blood concentrations oscillated too much on six than eight of our animals. We concluded that the doses varieties Braz. J. vet. Res. anim. Sci., São Paulo, v. 45, n. 2, p. 131-137, 2008 individually and the correct dosage as to important as the evaluation of the blood concentration of the drug and became viable for cell therapy

Fish, as poikilotherms, are subjected to the influence of the environmental temperature. It has been already reported that fish immune system is influenced by temperature; however, no information is available concerning if temperature has any effect on thrombocytes. Thrombocytes from fish reared at 6°C, 10oC and 20oC were assessed for alteration in their aggregation capacity. Thrombocyte percentages were altered by the water temperature at which the fish was reared, decreasing from 19% (at 10°C) to 13% (at 6°C) and increasing to 24% (at 20°C). However, the aggregation capacity was not significantly compromised by these temperature changes in an individual cell basis, which would suggest a capability of this cell, to maintain this indispensable function at different temperatures.


Os peixes, por serem animais pecilotermos, estão sujeitos à influências da temperatura do ambiente. Vários trabalhos já descreveram que o sistema imune é influenciado pela temperatura, contudo não existem informações concernentes à influencia desta sobre os trombócitos. Trombócitos provenientes de trutas arco-íris (Oncorrhynchus mykiss) mantidas a 6°C, 10ºC e 20ºC foram testados quanto à sua capacidade de agregação. A porcentagem de trombócitos sofreu alteração dependendo da temperatura da água em que os animais foram mantidos, diminuindo de 19% (a 10°C), para 13% (a 6°C) e aumentando para 24% (a 20°C). Entretanto, a capacidade de agregação individual de cada célula não foi significativamente afetada por essas mudanças de temperatura, o que sugere a qualidade dessa célula em manter essa função indispensável independente da temperatura.
